本研究利用同源克隆的方法从小麦中克隆到1个基因TaNAC,该基因编码的蛋白序列结构非常特殊,其N端为保守性差的NAC结构域,仅包含A,B,D3个亚结构域,C端序列与DREB家族转录因子相似性很高,且在进化关系上两者更近。干旱和高盐胁迫处理表达实验确认TaNAC基因受干旱诱导强烈表达,对高盐处理敏感。构建该基因的过量表达载体并通过农杆菌介导的叶盘转化法将TaNAC转入烟草,PEG胁迫处理实验表明转TaNAC基因烟草具有抗旱性,证明了TaNAC基因在烟草中成功表达,且提高了烟草的抗旱能力,可以作为烟草抗旱育种的优质候选基因。
TaNAC与已知的NAC转录因子同源性很低,同胁迫相关NAC转录因子的同源性也很低,从结构和进化关系看来,TaNAC所具有的抵御干旱胁迫的功能可能不是由NAC家族的共有结构特征决定的,其C端序列与DREB家族转录因子具有高度相似性,而DREB家族基因主要功能就是参与逆境胁迫应答,本实验所分离的TaNAC转录因子的功能是由N端的保守域决定,或是C端结构决定还需要更进一步的研究,这一工作将为研究植物转录因子提供新的思路。参考文献
