止颤汤对帕金森病大鼠神经干细胞诱导分化作用的研究

目的:探讨止颤汤对帕金森病大鼠神经干细胞的增殖及向多巴胺能神经元分化的作用;方法:实验动摘 要:
物分为3组,正常组及模型组用生理盐水灌胃,止颤汤组用止颤汤灌胃,分别在处理后7、14、28天免疫组化法检测,神经干细胞标志物nestin及其在脑内位置变化,多巴胺递质转运子DAT蛋白表达。比较各组差异;结果:在动物处理后28天,止颤汤组帕金森病模型大鼠脑内nestin及DAT数量较对照组增多,且多分布在中脑损伤区域;结论:止颤汤对帕金森病大鼠脑内神经干细胞的增殖及向多巴胺能神经元分化有一定促进作用。

Cell Differentiation of Parkinson's Disease Model Rats

Objective: To study the inducing effect of Zhichan Soup on the neuron stem cell Abstract:
differentiation of Parkinson's disease model rats; Methods: the rats were divided into three groups, the normal and model groups were administered with 0.9% normal saline by gavage ,and the Zhichan Soup group with Zhichan Soup,the nestin and DAT protein were checked with immunohistochemical method respectively after the rats were treated 7,14 and 28 days .Results: compared to the control group, the nestin and DAT protein were significantly increased in the Zhichan Soup group at the 28 days time point, and the nestin protein were mainly appeared in midbrain.Conclusion: Zhichan Soup has the effect to promote the proliferation and differentiation of the of neuron stem cell Parkinson's disease model rats.

神经干细胞(neuron stem cell,NSC),在疾病等应激的环境下,这些干细胞可被激活—增殖及向神经元分化,但这种作用很弱,难以再生出足够的神经元来修复受损的神经组织。如果通过外来因子的干预,促进体内神经干细胞大量增殖并向多巴胺能神经元分化,有望为帕金森病根本治疗提供有效方法。止颤汤在以往的研究中显示出较好的临床疗效,本研究观察了该药对帕金森病大鼠自身神经干细胞的增殖及向多巴胺能神经元的分化的作用。
1 材料与方法1.1 实验材料

1.2.1 PD大鼠模型制备 参照有关文献制作大鼠
大鼠用1%戊巴比妥钠(0.5mL/100g)腹PD模型[1]:
腔注射麻醉后,固定于立体定向仪上,用6-OHDA定向损毁大鼠单侧黑质多巴胺能神经元,损毁后2周,颈部皮下注射0.1%阿扑吗啡,每只0.2mL,检测大鼠向健侧旋转的圈数,大于7r/min,为模型成功,用于实验。
1.2.2 动物分组及处理 (1)正常对照组:大鼠经过抓取,固定,但不做手术,0.9%生理盐水4mL,每天分2次灌胃。(2)止颤汤组:制作大鼠PD模型,然后止颤汤1.2mL/100g灌胃(1.2mL/100g为以前动物实验中起效的最佳剂量)用9%生理盐水稀释为4mL,每天分2次灌胃。(3)模型对照组:模型制作后,大鼠0.9%生理盐水4mL,每天分2次灌胃。1.2.3 检测指标 分别于处理后7、14、28天处死动物。免疫组化检测时动物处死前1天,注射BrdU,每4h一次,注射3次。处死后经心脏注射肝素化生理盐水10min,然后用4%多聚甲醛灌注15min,取出脑
