ASCs而不是损伤区域周围角膜基质细胞的迁移。对照组缺损区周边新生胶原纤维直径比正常角膜直径略

大,我们推测这可能与缺损区周围蛋白聚糖含量的明显降低导致部分新生胶原产生融合有关。

本研究结果提示:以自体ASCs为种子细胞与可降解生物材料PLGA复合可以构建与正常角膜类似的组织工程角膜基质,维持角膜的透明度。我们猜测其可能的机制是角膜局部的微环境诱导ASCs分化成为角膜基质细胞,并分泌大量蛋白聚糖及胶原,从而形成具有正常功能的组织工程化角膜基质组织。而进一步明确细胞的来源以及是否发生诱导分化,尚需更深入的研究。

综上所述,以ASCs复合可降解材料可实现组织工程角膜基质构建并修复角膜基质缺损。ASCs来源广泛,取材方便,并可取自自体,有望成为组织工程角膜基质组织新的种子细胞来源。
